Tn/T Specific Agglutinin from Estuarine Crab Scylla Serrata with Potent Mitogenic Activity on Mouse Splenocytes and Antiproliferative Effect on Hepatocellular Carcinoma (HepG2) Cell     (13/May/2010)

A lectin was purified from the hemolymph of esturarine crab Scylla serrata by successive 40% (NH4)2SO4 precipitation, affinity chromatography on asialo fetuin- Sepharose column, Resource Q anion-exchanger in FPLC system and designated as scyllin-2. Scyllin-2 was a homogeneous monomeric protein of molecular mass 75 kD judged by SDS-PAGE and confirmed...

Effect of O-Glycosylation and Sialylation Inhibitors on Classical NLS-Dependent Nuclear Protein Import in HT29-MTX Human Colon Cancer Cells     (05/May/2010)

Glycosylation, including O-linked, mucin type, has an import role in the function and activity of many proteins, particularly those that are secreted or transcellular. Previous studies have indicated that mucin-type O-linked glycans may also be carried by intracellular proteins such as the stress-related protein Orp150 that is involved in classical...

Comprehensive Analysis of Sialyltransferases in Vertebrate Genomes     (17/Feb/2010)

Sialyltransferases are a subset of glycosyltransferases involved in the biosynthesis of sialylated glycolipids or/and glycoproteins. The aim of this review is to provide a comprehensive review and analysis of vertebrate sialyltransferase genes. Primary structure/ function relationships have been explored through the use of molecular phylogeny and phylogenomic approaches. Several animal...

Article: Peptide Mimetics of Terminal Sugars of Complex Glycans
Full-text PDFAbstract

Peptide Mimetics of Terminal Sugars of Complex Glycans     (16/Feb/2010)

In this project we asked whether short peptides would mimic complex carbohydrate structures and express biological activity. Amino acid sequences were identified as potential glycan mimetics by molecular modeling of docking to binding sites of lectins. The sequence HPSLK was synthesized as a quadravalent structure from a tri-lysine scaffold. Solid-phase assays showed that this peptide bound strongly to lectins from Triticus vulgaris (wheat germ agglutinin, WGA) and Dolichos biflorus (DB), which bind monosaccharides, and to...

Glycosaminoglycans in Human and Bovine Serum: Detection of Twenty-Four Heparan Sulfate and Chondroitin Sulfate Motifs Including a Novel Sialic Acid-Modified Chondroitin Sulfate Linkage Hexasaccharide     (09/Feb/2010)

Heterogeneous heparan sulfate and chondroitin sulfate glycosaminoglycan (GAG) polysaccharides are important compo- nents of blood circulation. Changes in GAG quantity and structure in blood have been indicated in cancers and other human diseases. However, GAG quantities and structures have not been fully characterized due to lack of robust and sensitive...

Identification of Chemically Sulfated/desulfated Glycosaminoglycans in Contaminated Heparins and Development of a Simple Assay for the Detection of Most Contaminants in Heparin     (02/Feb/2010)

Contaminated heparin was linked to at least 149 deaths and hundreds of adverse reactions. Published report indicates that heparin contaminants were a natural impurity, dermatan sulfate, and a contaminant, oversulfated chondroitin sulfate (OSCS). OSCS was assumed to derive from animal cartilage. By analyzing 26 contaminated heparin lots from different sources,...

Carbonic Anhydrase IX Shedding Through Exosomes in Renal Carcinoma Cells     (02/Dec/2009)

Carbonic anhydrase IX (CA IX) is a transmembrane glycoprotein highly expressed in clear cell carcinoma of the kidney and in a wide variety of other human cancers. Its expression is largely under the control of intratumoral hypoxia and in some subtypes of renal cell carcinoma (RCC), by the presence of...
